The extraction, isolation, structure identification and biological activities of chemical compounds of red alga Eucheuma gelatinae collected from the cultivate base in QiongHai city of HaiNan province have been studied. To the lectin, we studied its extraction, separation and purification. To the carrageenan, we studied its comprehensive extraction technological process. Three compounds (E1, E2, E3)have been obtained from Eucheuma gelatinae by isolation and purification means such as vacuum column chromatography, pressed column chromatography, thin layer chromatography, recrystallization, et al. The structures are determined by spectroscopic means including IR.~ æ…”NMR~. ?CNMR.~ DEPT~ æ…” - æ…” COSY. æ…” - ?C COSY~. MS. Compound E1 is Cholestane-5-ene-l, 3-diol, Compound E2 is Cholesterol, The structure of compound E3 is under presumption. Compound E3 shows anti-bacteria activity to in vitro anti-bacteria test. The extraction and separation condition of lectin was determined Pure lectin has been obtained by partial precipitation with 40?0% (NH4)2804 followed Sephadex G-75 gel filtration to exhibited a single band on SDS-PAGE. The lectin, designated as EGL, with molecular weight of 29,000 on SDS-PAGE. Hemagglutinating test revealed that EGL can hemagglutinates A, B, 0, AB four different type of human erythrocyte. The hemagglutinating activity is stable over a wide pH range and at a relatively high temperature. Eucheuma gelatinae contains large amounts of carrageenan. Aiming to comprehensive exploitation of Eucheuma gelatinae, We also studied the extraction of carrageenan in the residue obtained from the above-mentioned EGL research. The best extraction technological process has been determined by orthogonal experimental design. In additional, a fast, accurate and convenient content analysis method is developed for carrageenan.
