Tephrosia vogelii is a important insecticidal plant for having abundance rotenoid compounds. Among them, rotenone has the highest activity. It bring difficulties to rotenone extraction and separation from plants that rotenone decomposes easily in high temperature. Supercritical Fluid Extraction(SCFE) is a new technique developed lastly for chemicals extraction and separation. SCFE has vastly superiority to extract and separate natural compounds which are sensitive to temperature and difficult to do with common methods. The studies was proceeded on the the supercritical CO2 extraction technique of rotenone from Tephrosia vogelii. The extraction influences of technology conditions were systematically groped, The research results were listed as follows:1 The analysis method applied in this paper has been proved to be suitable to technology research of the studies on the SC-CO: extraction technique of rotenone from Tephrosia vogelii. 3 indexes, extraction ratio of rotenone. extraction ratio of total compounds and rotenone content percent in total compounds weight, were carried out with HPLC quantitatively determination combined with the sample weight loss after extraction. With these 3 indexes. The extraction influences of technology conditions were evaluated by synthesis. Then the best technology conditions were ascertained for different extraction aims.2 The pretreatment of rough material should be needed before extraction. Comminuting the material to 0.28mm size is most fit to SC-CO: extraction.3 Firstly static extracting 20 min then turn to dynamic doing is the better extraction mode than other ones.4 The orthogonal experiment was conduct with 3 factors and 5 levers. The studies shows that temperature is the most important factors. On extraction ratio of rotenone. pressure has a higher effect than CO: outflow quantity. While on extraction ratio of total compounds and rotenone content percent in total compounds weight, the latter has a higher effect than the former. When sample amount is 1 g, the best dynamic extraction conditions for rotenone extraction is 4000psi pressure. 45 癈 temperature and 30 mL CO:. Under this condition, the highest ratio of rotenone can meet 0.359% counted with engineering calculation. For total compounds extraction. 6000psi. 70癈. 40mL and the ratio is 10.06%. For rotenone content in total compounds. 3000psi. 35 癈. lOmL and the high content percent is 7.59%. So when conducting SC-CO: extraction of Tephrosia vogelii. the fittest conditions should be selected for different extraction aims.5 Adding modifier into extraction system has a substantial influence on SC-CO2 extraction effectiveness of Tephrosia vogelii. Methanol has been selected as the best one and the suit using coefficient percent is 10% occupancing in total fluid volume. Directly adding modifier into extraction system is better than pumping it during process.6 Contrasting with conventional methods. SC-CO: extraction Tephrosia vogelii has evident advantages: save in solvent, shorten extraction time, high extraction ratio, analysis precisely, nonpoison, nonpollution and so on. These features make it possible to industrially develop in future.7 The studies head to open the SCFE techniques in research field of nature products as pesticides. The research results can support industrial development of this technology and can be use for reference by the similar studies.
