Based on the heterogenetic reaction , by increasing the ratio of liquid and solid , the reaction of cellulose sulfation can be processed under the condition of stirring. The optimal conditions of reaction for preparation of sodium cellulose sulfate (NaCS) were found by the method of orthogonal design. Then the process was scaled up to 1L and 5L step by step. The comparison of the chemical and physical properties of NaCS prepared between in large-scale and in small scale showed that the scale up was successful. All three products can be used to encapsulate microorganism and cells grew well in different microcapsules prepared in three NaCSs.In the second pard of this paper, we studied NaCS-PDMDAAC microcapsule to culture bacillus subtilis which can produce nattokinase. We studied the effects of different concentration of carbon source and nitrogen source, and we also studied the effects of different seed volume and culture volume, and after several circuses of culture ,the total activity can be 2014UK/ml.In the third part of this paper, we studied SA/CS-CaCVPMCG Microcapsule to culture bacillus subtilis which can produce nattokinase. the crafts of making SA/CS-CaCl2/PMCG capsule was introduced.The effects of PMCG on the growth of bacillus subtilis and was studied. And we also studied the difference of microencapsulated cell and free cell on growth rate, consuming rate of xylose and producing rate of notokinase And we got high activity of enzyme after 6 batches culture. All the results show that the SA/CS-CaCl2/PMCG Microcapsule has good Biocompatibility.
