Many chiral drugs may differ in their pharmacological activities due to the difference of stereochemistry. Therefore, rapid and sensitive methods for the resolution and quantification of the individual optically pure enantiomers are of great importance; chromatographic methods on both the analytical and preparative scale have become of considerable interest in recent years. Particularly in the past 20 years, optical resolution by high-performance liquid chromatography (HPLC) has progressed rapidly, and has become a practical and useful method not only for determining optical purity but also for obtaining optical isomerso In this paper the separation of some enantiomeric basic durgs. insecticides (such as fenvalerate, triadimenol ) and sec-phenethyl alcohol has been investigated by means of chiral mobile phase additive (CMPA) or by theuse of chiral stationary phases (CSP) respectively.In part I , a review on chiral resolution by high-performance liquid chromatography(HPLC)is presented. Basically, two different approaches for the liquid chromatography resolution of enantiomers and the types of chiral selector are introduced. This review also deals with the development and present state of chiral selectors.In part II, the basic theory of separation by high-performance liquid chromatography ( HPLC ) and chiral recognition mechanisms for enantiomers are described.In part III to VI, ephedrine, tyrosine methyl ester, metoprolol, ibuprofen enantiomeric separation with chiral additive as mobile phase by high performance liquid chromatography are presented. Enantiomers of ephedrine, tyrosine methyl ester, metoprtlol, ibuprofen were completely resoloved with β -cyclodextrin or Hydroxypropyl- β - cyclodextrin as the chiral mobile phase additive on C8 column. In additional the influence of the mobile phase composition , pH% the mobile flow-rate , the concentration of chiral mobile phase additive on the separation factor are studied. The method with β -cyclodextrin, Hydroxypropyl- β -cyclodextrin as the chiral phase additive for the separation of ephedrine, tyrosine methyl ester, metoprolol, ibuprofen enantiomers have been establish.In part VII, the determination of metoprolol, tablets are introduced byHPLC on the Agilent Eclipse XDB-C8 column, with the mobile phase consisted of methanol-water(60:40). The average recovery of artemether was 100.5% with RSD of 1.56%(n = 5 ). The method is simple, rapid and the results is accurate and reliable.
