Lipstatin is an inhibitor of pancreatic lipases isolated from Streptomyces toxytricini, which tetrahydroderivative has been clinical used for treatment on severe obesity named Xenical.This paper dealt with the strain screening of Streptomyces toxytricini, the fermentation process optimization, the protoplast preparation and plasmid extraction .Streptomycin was used in strain screening combined with traditional mutation methods such as UV and NTG to screening Streptomyces toxytricini. Through careful study, we found that this is an effective method in strain improvement.The fermentation processing was optimized, the optimum fermentation temperature is 30℃, starting pH is 6.0, rotating speed is 220rpm. The optimum culture medium ingredients included: bean oil 5.0%, glycerol 2.0, defatted soybean powder 3.0, lecithin 1.6, MgSO4 0.005 % , ZnSO4 0.01 %.Successfully, we prepared and regenerated the protoplast of Streptomyces toxytricini. The parameters of making protoplast were studied. No plasmid was found in the strain we used..
