Based on rational selection and cultivation theory, at first, the article establishes a primary screening model of "Dephosphate adjustment". We carried out inducement to "the Cyclosporine A protoplast and screened on the plate with gradient phosphate concentration. The thus obtained mutant strain is further screened with a "final product resistance" model, and finally we obtained a mutant strain which is highly tolerant to high-concentration phosphate and final product.With relatively high concentration of phosphate formulation, the Cyclosporine A synthetic capacity of the selected mutant strain has achieved 2600 μ g/ml, being 80% higher than that of original strain. Besides, through optimization of seed cultivation, fermentation formulation, fermentation condition, etc., the capacity of the selected mutant strain has further achieved 3000 μ g/ml.We also have carried series of research on latter separation and purification process, with the process conditions like extracting, chromatographic, crystallization process, etc. studied. Thus the purification yield of Cyclosporine A has achieved 45%, 20% higher compared with the original yield. The spectra study shows that the purification and structure of the product is in compliance with those of USP reference.
