In this paper, through covalent linkage, aspirin and nicotinic acid were attached to 2-hydroxythyl methacrylate(HEMA) respectively, benzocaine and metronidazole were attached to mathacrylic acid respectively, thus unsaturated monomers A, B, C, D were synthesized. By using azobisisobutyronitrile(AIBN) as initiator, a series of polymeric carrier drugs were obtained through self-polymerization of these monomers and co-polymerization with MMA or other drug monomers. In order to improve the properties of these drugs, we also synthesized nano-drug-microspheres of nicotinic acid and benzocaine. These synthetic products were characterized with 1H-NMR,GPC and TEM.The results show as follows:(1)It is feasible to obtain polymeric carrier drugs by synthesizing unsaturated drug monomers firstly, then self-polymerizing and co-polymerizing them.(2)The composition of the copolymers can be controlled by adjusting the mol ratio of monomers attend the co-polymerization, so that polymeric drugs with various contents of drugs can be obtained.(3)Polymeric microspheres containing drug can be effectively synthesized by using emulsion polymerization.(4)TEM photos indicated that the diameters of drug nano-microspheres decrease with the amount of SDS and changed slightly with the amount of EtOH. So the diameter of microsphere can be controlled by adjusting the amounts of SDS or EtOH .These studies provided a reference for the synthesis of polymeric drugs and nano-drug-microspheres.
