In this article, the volatile components were extracted from the famous Chinese vinegar-Zhenjiang Vinegar using six different methods. Then these extractions were analyzed by Gas Chromatographic-Mass Spectrometric(GC-MS). It was shown that in these extractions there might be Ligustrazine which is regarded as a kind of hygienical substance. The method for Ligustrazine extraction from the Chinese medicine Ligusticum Chuanxiong Hort was simplified and improved. Using this improved method a kind of colorless and acicular crystal was obtained from Zhenjiang Vinegar. Compared to the retention time of standard sample using High Performance Liquid Chromatography(HPLC), the crystal was identified to be the Ligustrazine.The method of HPLC for the Ligustrazine quantitative analysis was established. The calibration curve was linear over a range from 30.7μg /ml to 153μg /ml. The exactitude, reappearance and recovery ratio of this method was tested by statistical analyse. Tracing the quantitative changes of Ligustrazine discovered that a little Ligustrazine firstly appeared during acetic fermentation. The quantity of Ligustrazine increase greatly after heating and hermetically storing. The quantitative changes of Ligustrazine, sugar and amino acid were analyzed in some vinegar samples. It was proved that Ligustrazine was come from Maillad action and the quantity of Ligustrazine was effected by fermentation level, production technics and storing time. So Ligustrazine could be used as quality criterion of Zhenjiang Vinegar.Ligustrazine is one of the active ingredients of Ligusticum Chuanxiong Hort. It was recorded that the content of Ligustrazine in Ligusticum Chuanxiong Hort is 180μg /g, although it is 138μg /ml in Zhenjiang Vinegar after 4 months storing and more than 700μg /ml after 8 years storing. It is shown that the health protection effect of Ligustrazine in Zhenjiang Vinegar can not be ignored.The Thin-Layer Chromatography(TLC) method for identifing Ligustrazine was also established. This method is fast, simple, convenient, less cost and can be used to analyze dozens of samples at one time. It is adapt to check Ligustrazine during vinegar production process.
