Nucleosides, which are the basic elements of nucleic acid, have crucial effects on the growth, development, propagation and heredity of almost all kinds of lives. They were mainly produced during the transcription. The abnormalism of inner-cell nucleosides may serve as biomarkers indicating potential health problems, which could be used in disease diagnosis. The nucleosides in natural are thought helpful in improving the immunity, and important in nutriology. Nucleosides in natural medicines were usually selected as quality control components.In this thesis, the analytic method of normal and/or modified nucleosides in human urine using micellar electrokinetic capillary chromatography (MEKC) was established. The operating parameters were optimized. This method provides a basic strategy in metabonomics study of human urine.Meanwhile, HPLC method was also established to analyze the normal and modified nucleosides in human urine. Furthermore, LC/MS/MS method was employed in identifications of fractions from LC separation.Cordyceps Sinensis is a kind of rare medicine, in which nucleosides are main effective components. Capillary zone electrophoresis (CZE) was employed to quantitatively analyze nucleosides in Cordyceps Sinensis. The differences in nucleosides between natural Cordyceps Sinensis and cultured Cordyceps mycelia were studied. CZE analysis results were also used to differentiate Cordyceps Sinensis from 6 different places in Qinghai province.
