In the first part of this thesis, the history of chemometrics and its research status are reviewed, The multivariate resolution and calibration as well as the three-way resolution and calibration in chemometrics were emphasized. It included the development of the trilinear decomposition algorithms as well as the applications of these algorithms to different fields of analytical chemistry, in particular, to three-way fluospectroscopy (excitation-emission matrix fluospectroscopy)In Chapter 2, a method of simultaneous determination of two main components of the Chinese traditional medicine cortex fraxini (aesculin and aesculetin) was introduced. The samples were measured by excitation-emission matrix fluospectroscopy (EEMs) and the data were resolved by alternating trilinear decomposition(ATLD) algorithm, This method don't need pre-separating the crude solution, and will determine these two components in the presence of the interference of other components.In Chapter 3, an excitation-emission matrix fluospectroscopy method combined with chemometrics method was developed to detect tetracycline(TC), oxytetracycline(OTC) , and methytetracycline(MTC), The alternating trilinear decomposition algorithm was used to resolve the seriously overlapped peaks of these three Pharmaceuticals. The results which obtained from the EEMs coupled with ATLD method were compared with those of HPLC-DAD, both results were very similar.In Chapter 4, a method was proposed for simultaneous determination of ofloxacin (OFL),norfloxacin (NOR) in the presence of enoxacin(ENO) by combining excitation-emission fluorescence with second-order calibration based on alternating trilinear decomposition method. The average recoveries of OFL and NOR were 100. 3 0. 52 (%) and 98. 1 0. 4 (%) respectively. In complex matrix, urine, the average recoveries of OFL and NOR were 100.7 0. 9 (%) and 101.4 4. 65 (%) respectively. The method was satisfactorily applied to the quantification of OFL and NOR in pharmaceutical preparations.In Chapter 5, tryptamine is the metabolic outcome of tryptophane, it coexist withtryptophane and tyrosine, its fluospectrum was very closed to that of tryptophane and tyrosine , it will disturb the detection of the tryptophane and tyrosine, In this chapter ATLD method was used to resolve the EEMs data of tyrosine, tryptophane under the interference of tryptamine, the results were satisfied.
