The safflower oil derived multi-unsaturated fatty acid monoglyceride was synthesized from net glycerin and virgin safflower oil catalyzed by inorganic bases. By orthogonal experimental design and macro-kinetics guidence, optimization of the preparation process was made. The optimum conditions are as follows: the ratio of glycerin to safflower oil is 3:1 (molar); the base catalyst content is 0.14% (on oil, weight); the temperature is 195; the reaction time is 70-80 minutes. Meanwhile, factors influencing the preparative reactions of monoglyceride yield were investigated. Experimental data showed there exist positive concentration effect. Furthermore, synerisms of NaOH:NaHCO3(1:1, molar) was observed. As a result of the above mentioned optimization job, high yield of monoglyceride (66.53%, weight) was acheieved, which was record high up to now. Finally, an optimized separation process was tried out, which featured with simple facilities, easy operation, less energy demand, and high efficency. The structures and properties of the substrate, intermediates, and products were characterized. In conclusion, a green process for the preparation and separation of safflower based monoglyceride was investigated, by reasonable design of the experiment, optimization focused on high yield of monoglycerid and stable of multi-unsaturation moieties on the fatty acid chain were carried out.
