The paper is made up of two parts, the first part is non gel capillary electrophoresis interaction analysis. Since migration time window is too small, capillary zone electrophoresis peak shift model can't exactly measure the interaction between drug and protein which have similar mobilities. The mechanism of molecular sieving of entangled polymer solution in non gel capillary electrophoresis (NGCE) was cited in the paper. In order to expand migration time window, entangled polymer was used to change the mobility of protein in the buffer solution. The model of interaction analysis between protein and ligand possing similar mobilities was established by NGCE. This method further developed the analytical methods of capillary electrophoresis interaction analysis.1. Since mobility ratio is independent of viscosities, it was introduced to the calculation model of capillary electrophoresis interaction analysis, in order to efficiently eliminate the effect of viscosities of entangled polymer solution. 2. Liner polyacrylamide was used as sieving polymer to determine the binding constant between chlorpheniramine and bovine serum albumin (BSA). In contrast with NGCE, capillary zone electrophoresis was also used to measure binding constant between chlorpheniramine and BSA. The results of two methods were almost consistent. 3. Dextran was used as sieving polymer to determine the binding constants between Indomethacin, and BSA, Irbesartan and BSA separately, which were similar with mobilities.The second part is chiral separation of capillary electrophoresis. The enantiomeric separations of chlorpheniramine and sibutramine by capillary electrophoresis using β-cyclodextrin (β-CD) as the chiral selector were studied.1. Binding constants between the enantiomers of chlorpheniramine and β-CD were studied at different pH. The relationship of resolution and binding constants was discussed. Since the concentration of β-CD at which the discrimination of enantiomeric mobilities was maximal was inconsistent with the concentration of β-CD at which the resolution of two enantiomers was optimal, the paper discovered the relationship of resolution and the mobility of two enantiomers.2. The enantiomeric separation of sibutramine by capillary electrophoresis using β-CD as the chiral selector was studied. The effects on enantioseparation were investigated and the two enantiomers could be separated well. Under the optimal operation conditions, the linear concentration region and the lowest detective concentration of sibutramine were studied. The quantitative analytical method of sibutramine and its enantiomers were be set up. Under operation conditions, the binding constants of two enantiomers and β-CD were studied.
