58 stains producing lipase were isolated from rich oil soil.The lipase from GXL02 has the highest activity to catalyze glycerolysis of oil. The strain GXL02 was identified as Acinetobacter sp.according to its character and 16S rDNA sequence analysis.The lipase production conditions of Acinetobacter sp.GXL02 was studied in detail.The lipase activity was 12.44u/mL after optimization of culture conditions.The results show that lipase production of GXL02 depended on fat and Mg2+, Ca2+ etc metal ion. Glucoce etc carbon source restrained lipase production.The optimal pH and temperature of the lipase for hydrolysis of olive were 9.0 and 50℃ respectively.The activity of lipase was no much change in 60℃ for 90min,and stable from pH3.0 to pH10.0.The lipase was used as catalyst to synthesize monoacylycerols in glycerolysis of palm oil in a solvent-free system.The result show that the optimal reaction temperature was 32 ℃.The optimal reaction condition would be :dosage of lipase 200u/g oil,water in glycerol 4%,pH of water 10.0,mole ratio of glycerol to oil 2.5 :1. After 4h,the content of monoacylglycerol in products was 41.42%.
