The main advantages of monolithic column are good hydrodynamiccharacteristic, low flow resistance as well as relative easy in preparation.A procedure for the preparation of methacrylate monolithic μ-HPLC columnsof 100 cm (eff) and 25 cm (eff) ×320μm i.d was described. The influence of thecontents of 1-propanol and the porogen solvent in the polymerization mixture onthe porosity and performance of the columns were investigated. Columns wereevaluated by an μ-HPLC system.A butylmethacrylate monolithic extraction column was used instead of theinjection loop as an on-line extraction unit. Compared to direct valve injection, thesensitivity was enhanced by 10~20-fold by using this unit.A two-step capillary isoelectric focusing and detection was developed forproteins separation. An electroosmotic pump (EOP) was used to migrate thefocused zones in the capillary through a detection zone. The separation and thereproducibility were better than the results obtained by using a mechanical HPLCpump.
