It is necessary to develop more effective and environment friendly inhibitors dueto the increased antibiotic resistance of bacteria. As one of the photosensizers,porphyrins show toxicity against microbes once activated by visible light. However,most porphyrins which have been gained are mixture. Thus it is valuable to producepure compound with antimicrobial activities.In this study, pheophrobide a was prepared by chemical hydrolysis ofchlorophylls. The molecular weight of pheophrobide a was 592.2 by LC-MS analysisand the UV-vis spectrum showed its absorption peaks at 410 nm and 663 nm.The pheophorbide a was found its efficacy in repressing the growth ofGram-positive bacteria such as Staphylococcus aureus while no obvious effect wasfound on the growth of Escherichia coli and its protoplast regenerationPurification of the pheophrobide a binding proteins was performed using aself-made affinity column. A membrane protein, whose molecular weight was about30 kDa, was achieved. The absorption peak of pheophrobide a was drifted due tothe binding of the protein .
