As an inelastic scattering, the scattering wavelength and the direction of the photons of Raman scattering changed with regard to those of incident light As it is known, the phenomenon that the Raman vibration of sample molecules absorbed on the rough surface of silver sol is obviously enhanced is named as Surface Enhanced Raman Scattering (SERS), therefore, the analytical method based on it Surface Enhanced Raman Spectroscopy (SERS).In this study, firstly, the elementary principle and progress of Raman and SERS were presented, particularly the recent development of its application in biological systems. Then, the following four systems were taken as examples to study the interaction between the little chemical molecule and biomacromolecule. Some interesting results were obtained, which displayed the advantage and potential of this technique for the research in biological systems.1. FT-Raman Spectra Study of Microscopic Damage of Rare Earth Ionsto the Space Structure of DNAThe character of microscopic damage of rare earth ions to the space structure of the calf thymus DNA was studied by FT-Raman spectroscopic technique. It was found that the Raman characteristic frequency and intensity of DNA molecule changed in different extent after dipped into rare earth ions solution. The results indicated mat phosphodiester groups of the backbone, deoxyribose and base of DNA were all damaged by rare earth ions. Concretely, the conformation and configuration of DNA obviously changed, including: hydrogen bond was broken, both single and double chains of DNA were disrupted and base pairs were damaged greatly.
