| Hyaluronic acid(HA) is one kind of acidic mucopolysaccharido which exists extensively in many tissues of people and animals.HA has many important physiological actions.So it is applied to such every fields as the medicines and cosmetics.But the main productive methodology of HA in our country still remains extraction method at present.It can't meet the increasing demand for HA day by day,so it is very essential to popularize the advanced technology of producing HA --fermentation method.Developing one downstream craft which can draw HA from the fermentation broth is the way by which fermentation method can be used in commercial production.Purpose : Designing a set of downstream extracting craft and technical parameters that can be used in commercial production.Making the HA product reach the quality requirements of cosmetic grade HA.Method:Using one kind of bacterias Streptococcus zooepidemicus,fermentation broth can be got by batch and supplementary food in fermentor.Studying the six steps of extracting:pretreatment,get rid of impurity, disengagement, purification ,dryness and grind. While studying every step,measuring the HA recovery rate, the loss rate of molecular weight, and the elimination rate of proteins.Choosing the best methods and conditions.Joining them into one route , and examine its result through the pilot test.Result: Had got a craft route and some technical parameters.In the pilot test, the HA product is white powder, the HA recovery rate is 70%, the molecular weight is higher than 1.4x 10~6, the content of protein is lower than 0.5%, the content of glucuronate is 39.1%.Analysing with infrared absorption spectroscopy and ultravioler spectroscopy,it can be seen that the products have reached the quality requirement of cosmetic grade HA.Conclusion: Getting a set of downstream extracting craft and technical parameters that can be used in the HA production process through fermentation.It has practical meaning for the fermentation method of HA production...
