Triptergium wilfordii Hook F. is a traditional Chinese medicine which is frequently used to treat the autoimmune and inflammatory disease, such as the rheumatoid arthritis (RA), skin disease, nephropathy and has shown great supplying foreground in organ graft. There are several medicines made from it in the market, such as multi-glycosides Triptergium wilfordii tablet which has antiphlogistic antalgic and antibacterial actions and has become the most commonly used-medicine for the rheumatoid arthritis (RA). Diterpenoid triepoxides are the important biologically active components in the plant, and among them triptolide (T) has been demonstrated to be the main active compound. However, multi-glycosides Triptergium wilfordii tablet is regarded as a toxic preparation and triptolide (T) has been found to be the most toxic among the active compounds, which is especially harmful for the liver, digest system, nerve system and procreant system. It can be seen that the toxicity of triptolide (T) has greatly limited the Chinese herbal drug to be applied widely. Another active compound tripchlorolide (T4) has also been separated from the grass and is thought a derivation of T (Lu, 1990). Its pharmacological experiments show that it also has antiphlogistic, antalgic and antibacterial actions but the toxicity of T4 is far lower than T and it is more hopeful to be used in larger scale than T. However, the amount of T4 is so much less than T that it is usually hard to be detected in the natural herbal grass and the raw material source is a big problem. There are four chapters in this thesis.Chapter one: a review on the research progression of the herbal drug, exploitable value and applicable foreground of Tripterygium wilfordii in recent years was introduced.Chapter two: an approach about the extraction, separation and determination of triptolide (T) which was the main active and toxic component in Tripterygium wilfordii has been established in this chapter.Chapter three: a new modification method in which the main active and toxiccomponent T was converted into tripchlorolide (T4) mostly has been established. The content changes of T in the herb and solid which were soaked by acidic water were investigated by reversed-phase high performance liquid chromatography.Chapter four: a micellar electrokinetic capillary chromatography (MEKC) method has been established for the determination of T4 in Chinese herb Tripterygium wilfordii Hook F. The influence of SDS concentrations, pH values of boric acid and borax buffer on the baseline separation of T and T4 in the herbal grass which has been modified by HCl-HAc were carried out. As the result, MEKC was found to exhibit good accuracy, precision and repeatability.
