Contamination of the surface and ground water by MTBE is not neglectable because of its wide use. In this paper, PM1, a pure strain, was used to degrade MTBE in aqueous solution in shaking-bottle. The optimal degradation conditions of MTBE were detemiined. Also the key factors which influenced biodegradation rate and the kinetic equation of biodegradation of MTBE were discussed by detecting the main intermediates.Above all, the analysis method of MTBE and its degradation products was established by gas chromatogram. The limit of detection of MTBE was 2 mg/L by DAI-GC/FID, and its degradation products were over 5 mg/L. The minimum detectability of MTBE was 0.04 μg/L by P&T-GC/MS-SIM, and its degradation products were less than 2.0 μg/L. Two methods were complementarity when used to detect different concentrations of MTBE.Optimal conditions of degradation of MTBE in shaking-bottle were as fellows: initial biomass was 50 mg cells/L, the temperature was 25 ℃, the pH was 7.2(neuter) and abundant oxygen.
