This article is divided into two parts: microbial transformation and study of antifungal agents. In the frist part, it was simply summarized that the application of microbial transformation and the two important reactions:side chain cleavage and hydroxylation of steroid respectively. In the study, the bio-transfomtion of canrenone with Aspergillus ochraceus was mainly done. AIM: To isolate the transformed compounds from the broth of Aspergillus ochraceus SEPI-4411. METHOD: (1)Selecting one strain SIPI-4411 from starting 4 fungal strains with potential ability in biotransformation;(2) Optimizing culture media of SIPI-4411 ;(3)Ensuring the factors which effected the transformation such as temperature, pH, aeration etc by single factor experiments;(4)Bio-transforming the canrenone with the enzyme system of Aspergillus ochraceus SIPI-4411; By solvent extraction and silica gel column chromatography, the compound SIPI-CANR was obtained. RESULT: By analyzing its MS, UV, NMR spectra data, and the chemical structure of SIPI-CANR was elucidated to be 15-hydroxylcanrenone. The transformation was in a yield of 60%~70% at the optimal condition and this finding is first reported so far.In the second part, it was studied for two strains SIPI-763 and SEPI-101 which have potential antifungal abilities. AIM: To isolate compounds from strains SIPI-763 and SIPI-101 with inhibitory activity against antibiotic-resistant. METHOD: Optimizing the fermentation condition and processing large-scale fermentation; using ethyl acetate extraction, silica gelcolumn chromatography etc, compounds were isolated from their broths. The partial spectra data of the compounds were determined. RESULT: By analyzing its MS, UV, NMR spectra data, the structure of one compound was identified to be identical with that of deoxy-toyocamycin.
