Polyamines exist in most living cells, They are biological polycations at physiological pH which are essential for cell growth and division. Their functions range from stabilization/modulation of membrane and mitochondrial activities to facilitating DNA transfection by phage and regulation of cell differentiation. Polyamines have a wide-ranging therapeutic potential ranging from applications in therapy of neurological diseases in development of new antidiarrheals in AIDS related cases and as anticancer agents. In the field of polymer science it has been of great interest to combine properties of polymers and useful functional groups. If amino-groups are incorporated into a polymer matrix. The polymer will be involved in a multitude of other important biological functions.We synthesized three unsaturated amino-monomers and three crosslinking reagents containing polyamine salts by reduction of azides, alkylation reaction and reduction of Schiff's Base. A series of amino-polymer and amino-copolymer have been synthesized by polymerization and copolymerization. Two series of hydrogels containing polyamine structure have been synthesized by crossing reaction with the crosslinking reagents.1. Synthesis and properties of amino-polymerIn this part, we synthesized three series of amino-polymer and amino-copolymer . Reaction of 4-vinylbenzyl chloride and sodium azide then polymerization and copolymerization with acrylamide produced a series of amino-polymer and amino-copolymers. Reaction of 1,4- dibromobutane and 4-vinylbenzyl chloride then copolymerization with methylacrylamide gave the copolymer containing polyamines. Reaction of 4-vinylbenzyl chloride and pyridylaldehyde then polymerization and copolymerization with acrylamideobtained the amino-polymer and amino-copolymer that contain pyridyl groups. The monomers and polymers were confirmed by 1H NMR, elemental analysis and IR . The properties of these polymer were researched by thermogravimetry analysis (TGA), UV-vis and potentiometric titration. The amino-polymer can form complexs with protein and Cu2+. The viscosity of these amino-polymer solutions were studied. The results show that the polymer solution accord with the common law of electrolyte solution.2. Synthesis and properties of crosslinking reagents containing polyamine and environment responding hydrogelsIn this part, A set of novel crosslinking reagents containing derivatives of putrescine are synthesized Reaction of 1,4-dibromobutane (or1,5-dibromopentane, 1,6-dibromohexane) and 4-vinylbenzyl chloride obtained the crosslinking reagents . The chemical structure of the crosslinking reagents were confirmed by 1H NMR and elemental analysis. The pH responding hydrogels were prepared by crossing reaction of the crosslinking reagent with acrylamide. The swelling ratio of the hydrogel were researched. The results show that the swelling ratio of the hydrogel decreased at lower or higher pH. Crossing reaction of the crosslinking reagent with N-isopropylacrylamide, the pH and temperature responding hydrogels were prepared. The swelling ratio of the hydrogel were researched. The results show that the swelling ratio of the hydrogel decreased at lower or higher pH. The LCST of the hydrogels were 33°C.
