Gardenia jasminoides Ellis is a commonly-used traditional Chinese herbal medicine with a wide range of pharmacological activities. Gardenia yellow pigment and geniposide are the two main chemical components in the fruit of gardenia Gardenia jasminoides Ellis. Gardenia yellow pigment ,with good water-solubility, had been widely used as food additive and industry dye. Geniposide is one of the functional components of hepatic protective and anti-inflammation activities. In order to develop useful techniques for producing of gardenia yellow pigment and geniposide with high quality, we carried out the following works: 1.Colorimetry and HPLC methods were established for the quantitative determination of gardenia yellow pigment and geniposide,the contents of the pigment and geniposide in the used sample of gardenia jasminoides Ellis were determined as 2.868% and 3.464%, respectively. 2.Gardenia yellow pigment was extracted from the fruit of gardenia jasminoides Ellis,and one of the components was isolated from the pigment, and was identified as crocin by mass spectrum(MS), NMR and infrared spectrum(IR). 3.Extracting conditions of gardenia yellow pigment and geniposide were obtained by orthonoal tests. The optimizing ratio of material and solvent, solvent concentration, temperature and extracting time determined as 1:12, 90%, 40℃ and 5h for gardenia yellow pigment , and 1:8, 70%, 80℃ and 5h for geniposide, respectively. 4.Macroporous adsorption resins were used to isolate and purify gardenia yellow pigment and geniposide. The adsorption characteristics of 13 kinds of resins for gardenia yellow pigment and geniposide were investigated, and HPD100A and H103 were chosen for their isolation and purification. 5. The procedures for the pigment and geniposide isolation and purification using HPD100A and H103 were established. Using these procedure, the colour value (E1cm1%,440nm)and O.D value(A238/A440,A325/A440) of gardenia yellow pigment obtained reached more than 360 and less than 0.24, respectively, and the purity of geniposide was more than 80%. 6. The small scale of gardenia yellow pigment isolation and purification procedure was successfully magnified . Extraction of 72g gardenia fruit was purified by 800mL HPD100A and H103 in turn, and the colour value (E1cm1%,440nm)and O.D value(A238/A440,A325/A440) of 0.63g gardenia yellow pigment obtained was egual to 360,0.206 and 0.201, respectively.
