In this paper, the advances of the molecular synthesis, properties and application of prophyrin and their metal complexes were reviewed, the application progress of porphyrin in diagnoses and therapy of tumors were specially expatiated. Bioactivities of nitric oxide and nitric oxide donor were also reviewed. The application prospect of metal complexes using as nitric oxide donor were anticipated. Four porphyrin ligands (a,b,c,d) were synthesised by different function groups —hydroxy,amino,carboxyl,methoxyl attaching to the phenyl groups on the meso-positions ring. The yields increased in the mixed of sovlents (propionic acid, acetic acid and nitro-benzene). Porphyrins ligand was converted into corresponding metal complexes on treatment with excess of M(OAc)2 (M=Co,Mn,Cu,Fe). The nitrosyl metalloporphyrin complexes were synthesized by metalloporphyrin complexe, heme and VB12 treating with nitric oxide. Using trifloracetic anhydride (TFAA) as condensing reagent, b can be activated to couple with biotin to from biotin–porphyrin conjugate 1b. 2b was synthesised by using1b treated with PdCl2. Using carbonyldiimidazode as condensing reagent biotin can be activated to couple with a compound to from 1a. 2a was synthesised by using 2a treated with Zn(OAc)2 the four compounds are never reported before. The structures of the above synthetic products were characterized by IR, MS, UV-vis, 1HNMR and elemental analysis. The spectra data of these new complexes, metal complexes were assigned. the spectra influence of different metal on the same ligand, or different ligands on the same metal were discussed and explained. The nitric oxide released process of nitrosyl metalloporphyrins in vitro was detected with diazotization assay at room temperature, the four nitrosyl metalloporphyrins(bCo-NO,bFe-NO,aCo-NO,aFe-NO)were good nitric oxide donors, the nitric oxide released efficiency is attained about seventy percent. These notrosyl metalloporphyrins are useful as NO donor drug. biotin-porphyrin conjugate is hoped to diagnose the early tumour as a stable biologic probe.
