Environmental hormone phthalate esters are an important kind of environmental hormone. It disturbs the biology which wants to maintain the balance in the body and adjust the production, the release, the shift, the metabolism, the response and the elimination of the normal hormone in the growth process, or causes the unhealthy influence and changes the endocrine function in the descendant biology which is not injured. In recent years, along with the industrial production and the use of the plastic product , the increase of the plastic trash massive, the phthalic acid ester enters the environment unceasingly, and becomes one of the most universal pollutants, existing in atmosphere, water , soil, organism and so on widely.At present , the main monitoring methods for phthalate esters are GC, HPLC and GC/MS. But the pre-processings of these methods are very complicated, time-consuming and costly. So it is necessary to develop a simple and quick monitoring method. Technique of immunoassay became one of the most competitive and challenging monitoring methods in the 21 century because of its quickness,sensitiveness, economy and wasting less samples.The investigation of literature shows that immunoassay is seldom used in monitoring environmental hormone, which means further research is promising and necessary. The purpose of this paper is to build a new immunoassay method for detecting environmental hormone phthalate esters. For this aim, I have done some researches as blow:1. The rabbits are immunised by the artificial antigens supplied by our seminar , and the antibodies of dicyclohexyl phthalate and dibutyl phthalate were picked up from the serum of rabbits and purified with DEAE-32 and ammonium sulfate solution.2. Microtiter plates are coated with antibody of dicyclohexyl phthalate . On the basis of sandwich ELISA, fluorescence immunoanalytical technique of dicyclohexyl phthalate was established with the signal of fluorescein isothiocyanate. This method has great advantages on sensitivity, selection and can be used in a broad range. Satisfactory results were gained when it was used to determine the concentration of dicyclohexyl phthalate in supply water and drinking water.3. On the basis of sandwich ELISA, fluorescence immunoanalytical technique of dibutyl phthalate was established with the signal of fluorescein isothiocyanate. This method has great advantages onsensitivity, selection and can be used in a broad range.
