The development of life science calls for improvement bio-chemical analysis. Polyer CD-like microfluidic platforms have many advantages for performing chemical and biological assays including low cost, high sample processing rates, esay operation, large parallel detection, minimum sample usage (usually in sub-microliter range), increased miniatutization, integration, and automation. There is much significance on research and development of polyer CD-like microfluidic platforms.According to demand for the task"studies on bio-chemical analysis based on polyer CD-like chips", the purpose of our work is to study on Chemiluminescence Immunoassay (CLIA) and Electrochemiluminescence assay (ECL) based on established platform of fabrication. This paper is organized as follows:In chapter 1, the status quo of research in the field of CD-like microfluidic platform, fabrication of organic glass microfluidic chip and CLIA were reviewed in detail.In chapter 2, firstly, a new method for fabrication of PMMA chip, namely elec-hot embossing was described. Secondly, some manufacture techniques of polyer chip such as hot embossing, CNC machining, thermal bonding were compared and concluded by the optimized experimental parameters. Finally, a PMMA CD-like microchip was finished successfully.In chapter 3, CLIA for HBsAg based on PMMA microchips was developed. We designed microchip system for CLIA in view of adsorptive ability of PMMA chips .In the experiment, there were such advantages as much less consumption of reagent (1/15 of the average), much shorter time (1/8 of the average). Further, the detection range is well coincident with the enzyme-linked immunosorbent assay (ELISA) result of Acon Biotech in Hangzhou city.In chapter 4, the electrochemiluminescence (ECL) behaviour of Luminol on micro- chip was studied systematically. It was obviously influenced by the electricparameters, oxygen content and acidity of the solution on 20 \iL micro-electrolytic cell chip. In the designed chip system, the result of research turned up trumps.
