The cadmium toxicity was revealed by many researchers. On the same time they found selenium can affect the cadmium toxicity. In this paper we thorough investigated the cadmium toxicity on primary cultures of rat Leydig cells. In addition, we also exposed cadmium and selenium combined to primary cultures of rat Leydig cells, assessd the antagonism of selenium to cadmium toxicity. The results were showed as follows:SD rats were applied to get the Leydig cells of testis, a multistep procedures (including dissociation of type I collagenase and Percoll gradient centrifugation) was employed. Results showed here: 90% Leydig cells were obtained. The purity of Leydig cells (more than 93%) was assessed through Wiebe stainng.Cadmium can affect on primary cultures of rat Leydig cells. Low or high dose cadmium can decrease the cells livability, the 3β-HSD activities of rat Leydig cells, the activities of SOD and increase the content of MDA. The activities of GSH-Px increased by low dose cadmium, and decreased by high dose cadmium.Compared with the group exposed by cadmium only, the index of the group exposed by cadmium and selenium depend on the dose of selenium. Low dose selenium can increase the livability of rat Leydig cells, the activities of SOD, the activities of GSH-Px, the 3β-HSD activities; and decrease the content of MDA. However, high dose selenium could decrease the livability of rat's Leydig cells, the activities of SOD, GSH-Px and the 3β-HSD were also decreased, and the content of MDA was increased.The experiment here revealed cadmium toxicity on primary cultures of Leydig cells of rat. In addition, we found low dose selenium have antagonistic effect to cadmium toxicity, however, high dose selenium can increase the toxicity of cadmium.
