Objective:From this experiment, we would be able to find a kind of method that can separate Anti-HBsAg Fab for industrialization production of Anti-HBsAg Fab to establish theory foundation in the future.Methods:1. Use of Streamline SP for selection and verification of optimal adsorption condition from E.coli preliminary separation and purification Anti-HBsAg Fab: The test tube method respectively determine that cation-exchange chromatographic media Streamline SP adsorb effect to target protein of preparation separation and purification at different pH and ion intensity of buffer. Then, use of 1ml Streamline SP prepacked column and 28ml Streamline SP self assembly column was run cation-exchange chromatography for verification adsorption effect.2.
