Redix Scutellariae, a common Chinese tradition medicine with assured pharmacological effect, is the dried root of Scutellaria Baicalensis Georgi . Baicalein and baicalin are two of the highest contents with appreciable pharmacological function in Scutellaria Baicalensis Georgi (Morita M et al., 2005).In this study , microbial transformation of Chinese traditional medicine—Redix Scutellariae was developed. An aspergillus niger was screened from soil with the capability of transformation baicalin to baicalein, determined by the thin lager chromatography (TLC) and high pressure liquid chromatograph (HPLC) method. The enzymes were investigated in-depth. The transformation between the substrate and the product was tracked by HPLC.In the further study the parameters of fermentation and medium were optimized, and the optimum medium and parameters of fermentation were obtained in the end. The optimum medium was as following: 10g powder of Scutellariae Redix, 2g NH4NO3, 0.5g ZnSO4 in 100ml water. The appropriate factors of fermentation was charge volume 26%, rotate speed 200rpm/min, natural pH, temperature 28-30℃, incubation time 6 days. Bioconversion rate was about 99%.The product of transformation was separated and purified through silica gel...
