Pentachlorophenol (PCP) was administered to activated sludge and aerobic granular sludge at different concentration to valuate wastewater performance of two kinds of sludge. Microbial composes and changes in sludge at different PCP concentration were monitored by T-RFLP technology, ammonia-oxidizing bacteria (AOB) were counted using Taqman real-time PCR for illustrating the mechanisms between sludge microbial components and their performances. Results are shown as follow:(1) PCP had more profound effect on ammonia nitrogen removal rate than that of COD, and with PCP concentration increase ammonia nitrogen removal rates decreased sharply. At lower concentration of aerobic granular sludge, ammonia nitrogen removal was effected more severely than that of activated sludge, while at higher aerobic granular sludge the result was on the contrary; SOURs of aerobic granular sludge and activated sludge were 16.7 and 50.1 mg O2/ (h·gMLSS), respectively, and half inhibit constants of PCP were 11mg/L and 8mg/L respectively; In effluent, nitrite nitrogen concentration from aerobic granular sludge SBR was far lower than that of activated sludge;(2) There were high similarity of microbial component between aerobic granular sludge and activated sludge, however the similarity decreased with PCP increase. Under PCP influence, microbial structures in two kinds of sludge changed gradually, and change of microbial component in aerobic granular sludge was more severely than that of activated sludge; According to the response to PCP, the bacteria could be categorized into four groups as follows:①The most sensitive organisms;②The more sensitive microorganisms;③The lower sensitive organisms;④The PCP-tolerant organisms, which become predominant within the sludge; Microbe which had similar response to PCP can be classified similar group in evolving branch;(3) With the increase of PCP concentration, the numbers of AOB in both types of sludge had no obvious change(p>0.05). The numbers of AOB had no obvious correlation with ammonia removal (p>0.05). The main effect of PCP on AOB in both types of sludge was to inhibit their metabolic activity;It was the most advantage of aerobic granular sludge that sludge can be maintained higher concentration in order to enhance ammonia nitrogen removal by heterobacteria;(4) T-RFLP combined with bio-information can be monitored microbe and its...
