Producing Of AflatoxinM₁ Kits And Raw Dairy Safety Alert Of AflatoxinM₁

Aflatoxin M1(AFM1) has strong toxicity and potential carcinogen. It's the high toxic metabolic product of Aspergillus flavus and Aspergillus parasiticus. AFM1 is the transform of Aflatoxin B1.It can be secreted in the dairy when the animals have intaken the foodstuff containing Aflatoxin B1. It is necessary to develop a sensitive, specific, quick and reliable detection method in order to monitor the content level of Aflatoxin M1.In order to detect the AFM1 monoclonal antibody, this text consulted many other documents, improved their methods, and prepared Aflatoxin M1 and OVA combination of the whole antigen.Depended on the hybridization tumour cell of antiAflatoxin M1 monoclonal antibody which Panwei had gotten, the subjuct was prepared ascites of mice, purified it, then got a great deal of antiaflatoxin M1 monoclonal antibodies, and finally established a competition ELISA method to detect the content of Aflatoxin M1. Based on this method, we prepared .the ELISA kits which can detect Aflatoxin M1 quickly and acutely.In many developed countries, the detection of Aflatoxin M1 has been well performed. However, the nationwide survey of Aflatoxin M1 had never been performed. In this research, the samples had been collected from the three main original cities of dairy production in China such as Shanghai, Shijiazhuang, Tianjin. 168 collected samples were detected by the kits which the laboratory made. The content of all samples measure up to the country's criterion, and two sample's were between 0.25ppb-0.5ppb,about 1.2% percent of all samples, the others were less than 0.25ppb.So it was advanced 0.25ppb to be the alert value of Aflatoxin M1. It not only accords with our actuality but favors to catching up to the developed countries.
