Bioactive proteins and peptides are a rapidly growing class of therapeuticagents for cure and precaution of disease with the development of biotechnology, but they show some disadvantages, such as short half-lives in vivo and proteolysis, etc. Preparation of chitosan microspheres containing therapeutic peptides and proteins has attracted more and more attention in recent years, due to their numerous advantages, such as improve the efficacy, ease pain or discomfort of the patients. Therefore, the objectives of this research are to develop a novel preparation method of SPG (Shirasu Porous Glass) membrane emulsification combined with chemical crosslinking process, which can prepare biodrug-loaded microspheres with high bioactivity preservation, high entrapment efficiency, and stable release behavior.First, the modification condition of modifying hydrophilic SPG membrane into hydrophobicity was invested. SPG membrane must be modified to be hydrophobic because hydrophilic membrane is unsuitable for preparation of uniform-sized W/O emulsion. The KP-18C (a silane coupling agent with a C18 hydrophobic chain) was chosen as the modifier after the optimization of modification conditions.
