Persistent Toxic Substances (PTSs) can resist photolitic, chemical and biological degradation, therefore due to their persistence they show ubiquitous distribution in the environment. Long-range transport can occur mainly by advection, even at global scale, through the atmosphere and freshwater and oceanic currents. Some of these substances are characterized by highly hydrophobicity and can bioaccumulate in living organisms up to levels that may impair normal biological functions and disturb the maintenance of blood hormone's normal level, and affect the human bio-system such as reproduction and developmental disorder, tumors occurrence, neurosystem, and immune system. Biomagnification through food webs has also been observed.At present, the attention to PTSs is enlarge from natural environment and wild animals to human health. We have observed the neurocyte toxic effect of the predominant contamination-Dibutylphthalate (DBP) in Three Gorges from the molecular and cellular level, and penetrated into case-control study to observe the correlation between environmental agents and cognitive impairment of pre-school children. It maybe offers scientific evidence to promote the human health.Methods:(1) Lab works: PC12 cells were incubated in culture media with5,12.5,25,50,100, 200 mg/L Dibutylphthalate for 24, 48, 72 and 96 hours respectively. Cell Counting Kit-8 (CCK-8) test, Trypan blue exclusion test, Lactate dehydrogenase (LDH) release ,glutathione peroxidase (GSH-Px)activity,malondialdehyd(eMDA)concentration of PC12 cells, DNA gel electrophoresis were performed to detect proliferation, cell membrane impairment, and apoptosis of PC12 cells, and RT-PCR were performed to detect the regulation effect for the apoptosis related gene such as P53, Bax and Bcl-2.
