Nano-biosensing is the most execiting cross subject, including nanobiology, nanoelectronics and nanomaterial science. Application of gold nanoparticles is one of the most important research contents in protein detection. Piezoelectric TSM immunosensing technique, which incorporates high sensitivity of piezoelectric response and high specificity of antibody-antigen immunoreaction, is sensitive to multiple signals such as the surface mass, and the density, viscosity, conductivity, and dielectric effect of the test solution. It may present some outstanding advantages including desirable simplicity, high sensitivity, rapid response, wide detection range, effective cost, real-time digital output, and free from tracer labeling and sample purification etc. This kind technique shows an attractive future of potential applications in the biochemical diagnosis of clinical diseases.In this paper, we have constructed piezoelectric biosensing real-time analyzer, biotin-streptavidin system was adopted as a test model to reflect the piezoelectric biosensing real-time response characteristics; gold nanoparticles and immuno- nanoprobes were prepared; based on the immunoagglutination theory and the non-mass piezo-sensing effects, a novel pize-immunosensing system for protein detection has been proposed. The present thesis is composed of the following three parts:(1) Because there are strong affinity and high specificity between biotin and streptavidin, biotin-streptavidin system was adopted as a test model to reflect the piezoelectrica biosensing real-time response characteristics. The adsorption deeds of biotin-streptavidin system were researched in this paper. Effects of pH on adsorption deeds of biotin and streptavidin, Impacts of experiment condition on reaction of biotin and streptavidin including incubating time and concentration of streptavidin and reaction time, and reaction characteristics were discussed. It was demonstrated that the adsorption of streptavidin is the best after incubating 8 hours in concentration of 100μg/ml when pH is 7.4. Under the optimal experimental condition, the frequency shift is linear with the biotin mass concentration in the range of 0.11~1.65μg/ml.(2) Gold nanoparticle suspensions were synthesized by reduction of HAuCl4 solution with trisodium citrate. By analysis of TEM, it was shown that gold nanoparticles are rotundity and equirotal, and average diameter is 13.8 nanometers.
