Photodynamic Therapy (PDT ) has been an available technique for the treatment of Cancer. Compared with traditional cancer therapies ,such as surgery, chemotherapy and radiation therapy, PDT has many excellences such as without destroying healthy tissue, no suffering and little by-effect etc. Hematoporphyrin derivative (HPD) is the first photosensitizer widely employed for clinical use with efficient therapy result, but also with a series of disadvantages such as compositional variability, weak absorption in red light region and high photosensitive toxicity. Phthalocyanines possess excellent physical and chemical properties and have become one kind of promising PDT agents to replace HPD.Coumarin derivatives have anticancer, antibacterial and biological activities, thus we try to introduce coumarin groups onto phthalocyanine rings, in order to increase the PDT activity and bioavailability of the phthalocyanines. In this report the main results are summarized as the follows:1, Five precursors with coumarin group for the synthesis of metallophthalo -cyanines substituted with coumarinoxy group were obtained, the precursors are 4-(7-coumarinoxy)phthalonitrile,4-(7-coumarinoxy)phthalonitrile,3-(4-methyl-7-cou-marinoxy)phthalonitrile,4-(4-methyl-7-coumarinoxy)phthalonitrile,4,5-(4-methyl-7-coumarinoxy)phthalonitrile. They were characterized by IR, 1HNMR , ESI-MS and Element analysis;2 , A series of metallophthalocyanine complexes α-(CMO)4PcM, β-(CMO)4PcM,α-(MeCMO)4PcM and β-(MeCMO)4PcM (M=Zn(II), Cu(II), Co(II), Ni(II); CMO is 7-coumarinoxy group; MeCMO is 4-methyl-7- coumarinoxy group) were obtained by using the above five precursors, and they were characterized by elemental analysis, IR, UV-Vis and ESI-MS.3, The influence of the substituted groups, the sites of the groups on the Pc rings , the central metal ions and the solution on the absorption spectra and the fluorescence property of the metallophthalocyanine complexes was investigated and discussed.
