In this study, matrine(MAT) was taken as the model drug and loaded by polybutylcyanoacrylate(PBCA) to prepare Matrine Polybutylcyanoacrylate Nanoparticles(MAT-PBCA-NP).The formulation and preparation technology of MAT-PBCA-NP colloidal solution and its lyophilization injection were systematically studied. The release kinetics in vitro,characteristics of distribution and pharmacokinetics in vivo were examined.The particle size, particle distribution, associating ratio(AR) and drug loading(DL) were employed as criterious to evaluate the influence of different preparation technology, pH, temperature etc. On the properties of MAT-PBCA-NP by single-factor trial and the main influence factors were taken out. Uniform design method was applied to optimize the fonnulation and preparation technology.The encapsulation rate of MAT-PBCA-NP after optimization was(84.91±1.76)%, the drug loading was (16.98±0.35)% and the diameter is (157.4±22.4)nm.The formulation of MAT-PBCA-NP lyophilization injection was established based on the criterious of its appearance,color and dispersing properties. As a result, the problems that the colloidal solution has poor physical and chemical stabilities and its difficult to disperse in water were resolved. The stability of MAT-PBCA-NP was not affected by the lyophilization process.
