Study On The Active Peptide From Jellyfish And Its Effect Of Decreasing Blood Pressure

The common component and the composition of protein of the umbrella and oral arms of fresh Jellyfish are detected and analyzed in this article. The protein of Jellyfish is the half of the whole dry weight and includes kinds of amino acids and abundant necessary amino acids. Acidic protease. alkaline protease. neutral protease. trypsin . pepsin and the compounding of two of them were used to hydrolyze jellyfish, respectively. According to inhibitory activity against angiotensin I -Converting Enzyme (ACE) of hydrolysates and economic benefit alkaline protease was selected for the proper enzymes to prepare ACE inhibitory peptides. In order to look for the optimal hydrolysis conditions of alkaline protease , orthogonal experiment were processed. After analyzing the inhibitory activity against angiotensin Ⅰ-Converting enzyme (ACE) of hydrolysates. the optimal hydrolysis conditions were ascertained, ratio of enzyme and substrate is 2%,the optimal hydrolysis temperature is 50℃, the optimal pH is 9,concentration of substrate is 4% . And the hydrolysis ratio is 41.8%, acid-soluble protein is 31.3mg/ml.moreover, the inhibitory activity against angiotensin Ⅰ-Converting enzyme(ACE)of hydrolysates holds the line though without bitter seasoning and sterilization.The jellyfish hydrolysates was selected and purified by the ultrafiltration membrane molecular-weight cut-off 2500 and 250Da,it is found that high active fraction with ACE inhibitory activity was the peptides which have a molecular-weight among 250 and 2500.Then the high active fraction was separated by Sephedex G— 15 columniation ,gained four fractions ,two of which have high active fraction with ACE inhibitory activity.
