The most important problem on using bio-strengthening technology in the process of sewage treatment system is engineering bacterium leaking, pellet is a new type of biological carrier immobilizing microbe, it offer the new method and road for this problem. Pellet is a kind of bio-granule that can form spontaneously in the process of ferment. It has a good biological activity, fast sedimentation velocity, good condition of liquid solid separation. So on a proper environment, pellet can form spontaneously. There is no report on using pellet as a bio-carrier immobilizing bacterium at the home and broad. It may be lead to a reformation on the field of biological fixation.This paper paies more attention on mechinics and the method of reproduction, optimize culture medium and condition of culture of aspergillus. By optimizing the culture medium and condition of culture, We ascertain that optimum carbon source is sucrose, and optimum nitrogen source is ammonium chloride,the important microelement are Fe2+,Mg2+. When the age of spore is 26d, the inoculum concentration is 104per litter, the culture temperature is 37℃, the initial culture pH is 5, the revolution of shaking table is 160rpm, the percentage composition of CaCl2 is 0.02%, the percentage composition of tween-80 is 0.5%, pellet will be at the best station and good decolouration.At the same time, this paper has some research on the formation mechinics and the method of reproduction, we bring forword the conception of saturation nuclear, explain the formation mechinics of pellet at the early stage growth by this conception, we recorded the change of structure and shape of pellet systematic by paraffin method at first time. The process of pellet formation can be separated into three stages: the single spore or spore nuclues sprout and mycelium outgrow; mycelium grow further, and at the action of external force mycelium form pellet; as mycelium regrow, pellet autolyze for the mal-condition of culture. Experiment verified the feasibility of reproduction using mycelium fragment,and we grind pellet at 1800rpm for 30 seconds, and can get the best degree of grinding of pellet what is optimal for reproduction.
