Based on the structure of cell wall, cellulase was employed to improve the extracting rate of effective components in herbs. In the cell wall of herb which is like a barrier to prevent the effective component from dissolving efficiently. Cellulose which isn't soluable in water or some general organic solution ccupies a large part of it. However, the insoluable cellulose can be hydrolised by cellulase into soluable components, the bad influence of cellulose can be eliminated through the addition of cellulase in this way. The concents of cellulose in dry pingbei, pasonia lactiflra Pall and Paeonia suffruticosa Andr are 35.63%, 30.92%and 30.98% respectively.Taking alkaloid as targeted effectibve compenents, the cellulase was added to the traditional extraction process pinbei. Through sigle-factor and orthogonal test, the extracting efficiency of alkaloid from pingbei was highest at 60℃; pH:4.5; cellulase amount:0.25g;hydrolysing time:3.5h, herb amount: 20g. Compared with the traditional process, the extracting rate increased by 39.2% from 0.0765% to 0.1065%.Taking paeoniflorin as targeted effectibve compenents, the same enzyme was applied pasonia lactiflra Pall every time. As for paeoniflorin in pasonia lactiflra Pall , the highest extractiong rate of 4.15% was obtained at 40℃; pH:4.5; cellulase amount:0.15g;hydrolysis time:3.5h, herb amount: 20g .Compared with the traditional process, the extracting rate increased by 6.81%.Looking up extrodinary amout of technique literatres , the extracting rate of paeoniflorin in pasonia lactiflra Pall was the highest.Many factors affect the extracting process using enzyme, so not every herb is fit for the new method such as the Paeonia suffruticosa Andr. Extracting rate of paeonol in it decreced in this experiment. It is likely the result of some large amout of inhibitors such as tannin, semi-cellulose and some metal ions and so on.The application of cellulase in herbs are not very profound currently. In this experiment , pinbei was taken as the substrate in this experiment , the optimized enzyme-assisted extraction process condition was derermined based on the extracting rate of alkoliosd and the optimized hydrolyzing condition was settled synchronously based on the contents of glucose.The corresponding conditions including temperature, pH, reacting time and enzyme amount were same by and large for pinbei. The kinetics character between the residual cellulose and initial cellulose at certain time wasinvestigated.
