This experiment is on the base of engineering bacterium fermentation and primary extraction, use two different kind of chromatography way to extract the recombinant human interferon a2b, the sticking point is exploring non McAb.During the process of purification in gene engineering production, all through, the affinity chromatography use as a high efficiency and handy separation technique, use this method to purify have the good specificity and the purity of the production is very high. But the price is expensive,low capacity and difficult to treat, because of its washing condition is rigor,so its yield is only within touch 60-70% of other method. In addition, as the result of easy to contaminate, so usually is not as the first choose use for develop technique. So that in this work we choose another kind of affinity ligand, use IMAC to substitute the traditional McAb affinity chromatography. According this experiment result, the purity of the sample protein after chromatography can achieve more than 95.0%, the specific activity can achieve more than 1.0 X 10~8 IU/mg, the sample after final chromatography of experiment group is more better than the reference group on the purity of electrophoresis and specific activity, it accord with the《Chinese pharmacopoeia》Ⅲpart standard, but the final yield, the total activity of three sample of experiment group isl.4times as the reference group's, so that's means use IMAC to purify recombinant human interferon a2b increase the amount of IFN in 40%, it can use for a great deal of production. The introduce of the IMAC column increase the unit yield and productivity, it has the good economy efficiency and social efficiency.
