Along with the development of industry and agriculture ,more and more surfactants are applied to production, such as detergents and chemical spray, which result in serious environmental pollution. As a most commonly used anionic surfactant, sodium dodecyl sulfate play an important role in the environment protection. Utilizing microorganisms to degrade contaminants is an effective measure and can not result in the second pollution to the surrounding environment. Therefore, the separation of degradative bacterium and the study on degradative characteristics hold great significance.A bacterial strain PSE-1 with the ability to utilize sodium dodecyl sulfate(SDS) as the sole carbon source was isolated from the soils polluted by SDS and identified as pseudomonas sp.by sequential analysis of 16SrDNA. In order to promote the SDS degrading abilities of the bacterial strain PSE-1, SDS is used as inducing chemical to acclimate the bacteria. The result showed that the SDS degrading abilities are greatly promoted from 30mg/l to 400mg/l. Meanwhile, this paper focused on the study of the biodegradation characteristics of SDS under different condition by strain PSE-1. The results showed that the PSE-1 can effectively removed SDS. At the same time, a plasmid strip was found from strain PSE-1 through plasmid detection. The experiment of plasmid elimination conformed that the plasmid has some relation to the ability of SDS degradation which is a degradative plasmid. The study on degradative characteristics and the discovery of degradative plasmid play an important role in cloning genes determining SDS biodegradation and constructing engineering strain which can degrade many kinds pollutions.Methylene-blue colorimetry of determining anionic detergents in water is, at present, a commonly used measure. But this method need excessive pharmaceutical, cost much time and require extracting repeatedly. This article makes a modification of the national standard method. The result shows that the improved method can avoid the shortcoming and have some advantages such as operating rapidly and simply, having good analytical correction and precision and low analysis cost and the results can meet the needs of environment emergency monitoring.
