Cyclodextrins are a serics of cyclic oligosaccharides consisting several glucose units linked throughα-1-4-linkages,α-β- andγ- cyclodextrins, these which consist 6, 7 and 8 glucose units respectively, are the most common ones. Cyclodextrins and their derivatives can selectively bind guest molecules to from complexes with their hydrophobic cavities. In this way, many properties of the guest compounds are greatly improved such as solubility and stability.β-cyclodextrin (β-CD) and its derivatives, based on their special structures and abilities, have been widely studied in many fields such as drug modification and solubilization, molecular recognition and identification of chiral reagents. This thesis including the following main points on synthesis technology ofβ-cyclodextrin derivatives and study on their performances:1. Using sodium hydroxide base as catalyst, different substitution degree hydroxypropyl-β-cyclodextrin was synthesized through the reaction betweenβ-cyclodextrin and propylene oxide. The influence of reaction time, temperature, reactant ratio and density of sodium hydroxide on the substitution degree of hydroxypropyl-β-cyclodextrin was discussed, which gave a yiele of over 40% and a substitution degree of 4~9. Commutative ion resin of strong acid instead of hydrochloric acid neutralized sodium hydroxide meliorated step of subsequent treatment. Study on amplifying experiment synthesis technology of hydroxypropyl-β-cyclodextrin.2. Model in determination of hydroxypropyl in 2005 China pharmacopoeia appendix VDF, the substitution degree of hydroxypropyl-β-cyclodextrin was assayed (Determination result of the method is called DSchemical approch)- Study on Relationship between DS chemical approch and determination result of 1H-NMR (this is called DSnmr)·Invent a simple method of accounting DSnmr by DSchemical approch·3. Study on inclusive performance of acetyl salicylic acid/HP-β-CD andβ-carotene/HP-β-CD. Assay inclusive ratio of inclusive compounds using UV-spectrum. Exam stability of acetyl salicylic acid/HP-β-CD in water of pH=7.0, sodium bicarbonate solution of pH=7.95, 9.10. The result indicated inclusive compound was not photolysis within 1h in these solution; Study on photestability ofβ-carotene/HP-β-CD under natural illumination, This experiment indicated this inclusion compound not to have photolysis in 4 days.4. Polymer-ethyloic-β-CD was synthesized by means of carboxymethylation then polymerization and polymerization next carboxymethylation separately. Study on absorbability of fuchsine onβ-CD, polymer-β-CD, and these two methods obtain polymer-ethyloic-β-CD. The absorbability of polymer-ethyloic-β-CD on fuchsine was the best obtained by the way polymerization first and then carboxymethylation, reached up to 49.03×10-4g·g-1.
