China possesses a wealth of eggs about product and consumption. Along with the increasing consumption and centralized processing of egg, a large number of discarded eggshells make pollution. Now, the comprehensive utilization of eggshells becomes an urgent social problem. The applications of eggshell had been studied at abroad and in China, but how to use eggshell membrane efficiently is still lack of research. Eggshell membrane is potential resource of protein and peptide which has a very considerable value. Accordingly, how to use eggshell membrane is discussed in this paper systematically.Firstly, the key step, the new technology of extracting biologically active peptides by enzymatic hydrolysis from eggshell membrane is studied. Compared with traditional process of the acid or alkaline hydrolysis of proteins, it has a simple technology, mild condition, scarcely destroying peptides and could keep nutritive value. In addition, it won't cause pollution to the environment. The technology has a good environmental benefits, social benefits and promotion prospects.In the process of studying enzymatic hydrolytic conditions, this paper brought forward Response Surface Methodology (RSM) firstly. The effects of main factors were discussed and achieved the optimum reaction condition through monofactor and Response Surface Methodology (RSM) test. The results are as follows: the value of pH is 9.6, enzymatic hydrolytic time is 2.04h, enzyme dosage is 3.9% ,temperature is 67℃, and solid-liquid ratio is 1:15. Furthermore by the response surface test, the value of pH and the enzymatic hydrolytic temperature have notable effect on DH and content of bioactive peptides, enzyme dosage and enzymatic hydrolytic time and solid-liquid ratio have little effect.Secondly, hydrolyzed polypeptides were purified with reversed phase high performance liquid chromatography (RP-HPLC).The Chromatographic conditions during the separation and purification were studied and optimized in this thesis, and it is helpful for further studies.
