Wild wine yeast is indigenous yeasts which occurs in native vine and natural circumstance. Native yeasts have some unique characteristics of metabolize which can bring special flavor for wine in their mirco- environments. It has been a hot topic to study wild wine yeast in home and oversea.Xianjing Province is the largest viticultural area and a new district for wine production, which own the unique climate and geography circumstance. The aim of this study is to find out species, distribution and character of wild wine yeast in Xinjing, and to build wine-related yeasts collection about Xinjiang area. These studies are profitable to provide supports for screening, breeding of industry yeast. Meanwhile they are very significant to prevent wine from homogenous.In this study, 426 strains were isolated from Manasi district and Shanshan district of Xinjing. The collected yeasts have been classified and identified by biochemical methods and physiological characteristics, WL medium was used to profile the strains.The restriction patterns analysis generated from the region spanning of the internal transcribed spacers (ITS1 and ITS2) and the 5.8SrRNA gene with four restriction endonuclease(Haeâ…¢,Hin6â… ,Hinfâ… ,Aluâ… )was performed together with the analysis of 5.8SrRNA gene sequence. Species, distribution and characteristics of these yeasts were reported for the frist time of Xinjing area.Seven genus and ten yeast species were identified in this study, including:Candida zemplinina,Candida stellata,Cryptococcus flavescens,Hanseniaspora uvarum,Issatchenkia orientalis,Issatchenkia terricola,Pichia kluyver,Rhodotorula graminis,Saccharomyces dairencnsi and Saccharomyces cerevisia.Hanseniaspora uvarum,Issatchenkia orientalis,Pichia kluyveri,Rhodotorula graminis,Cryptococcus flavescens,Saccharcomyces ceresive are the species isolated from Manasi district.The strains isolated from Shanshan district belong to six species:Hanseniaspora uvarum,Saccharcomyces ceresive,Pichia kluyveri,Issatchenkia terricola,Candida zemplini-na,and Candida stellata.Hanseniaspora uvarum is the mainly species in spontaneous fermentation, inhabited of 80.81%, and other is Pichia kluyveri,7.69%; Issatchenkia orientalis,4.80%;Rhodotorula graminis,2.4%;Cryptococcus flavescens,2.4%;Saccharcomyces ceresive,1.9% in Manasi district. While Shanshan district Hanseniaspora uvarum inhabited 88.09%, Saccharcomyces ceresive,5.5%;Pichia kluyveri,2.75%;Issatchenkia terricola,2.29%;Candida Zemplinina& Candida stellata 3.66%.
