To prevent peridural adhesions after lumbar laminectomy, meloxicam-loaded chitosan membrane was prepared and evaluated in vivo.Chitosan was chosen as membrane materials and the conditions of preparing membrane with the four different kinds of chitosan were studied. The optimum conditions were found. The concentration of acetum was 2.0%, the concentration of NaOH was 2.0%, the drying temperature was 50℃, the mass rate of glycerol and chitosan was 2.5:1, the concentration of chitosan solution was 11% (20kDa), 9% (47kDa), 6% (100kDa), 4% (280kDa), the vloume ratio of different chitosan solution was l:2(20kDa and 280kDa), l:l(47kDa and 280kDa), 3:2(100kDa and 280kDa).After the antibiotic activity, dilatability and degradability of membrane being determined, the mixed membrane with the chitosan of 47kDa and 280 kDa was chosen as the carrier with drug.The ultrastructure of chitosan membrane was observed. The results showed that the structure was closed and the surface was flat and smooth in vacant chitosan membrane and there was no obvious apart on the cross section in two-double membrane. Tenuous porous structure could be found in chitosan membrane from high micrograph. Mechanism of membrane hole was the medicine melt to porous structure in meloxicam-loaded chitosan membrane.The single and two-double meloxicam-loaded chitosan membrane were made and those release capability in vitro was studied. The laminectomies were performed with healthy female SD rats at each lumbar vertebrae L2. Two-double meloxicam-loaded chitosan membrane was placed over the defect. The result showed that the meloxicam-loaded chitosan membrane could prevent the peridural adhesions to better extent.
