The Raman Spectroscopy is an important tool for researching the structure of the lattice or molecule.It can provide the plenty of information about the inherent frequency,the symmetry and the inner interaction of molecular vibration,behavior and the reaction of the interface.The normal Raman signals is inherently weak,thus preventing the possibility of achieving low detection limit with normal Raman spectroscopy.In order to obtain Raman spectra with higher quality and lower inerference,people have developed a series technology of Raman spectroscopy:Laser Raman spectroscopy,FT-Raman spectroscopy,Surface enhanced Raman Scattering(SERS),Resonance Raman spectroscopy,Time-Resoled Raman spectroscopy and so on.SERS,as its high sensitivity,high resolved,no water and fluorescence interference,good stability,has been used more and more widely,for example,in interface field,the state of the absorbates on the substrate,the orientation and configuration of large bio-molecule and so on.This paper researched the Raman and SERS spectroscopy of some bio-molecules contained carboxyl,proteinic amino acids,DNA,anticancer medicine and shikimic acid.According to the SERS rules and mechanism,the adsorption and reactive manner on the substrate of Ag nanoparticle and core-shell Au/Ag nanoparticles are speculated,also the effective factors involved concentration and pH values are discussed.Each part list in the fellow:1.The Study of FT-Raman Spectroscopies of side chain amino acids,and Surface Enhanced Raman Scattering Adsorbed on Core-Shell Au/Ag NanoparticlesThe Raman spectroscopies and SERS of side chain amino acids(containing leucine,isoleucine and valine)on Au/Ag alloy colloids are obtained and interpreted.The Raman peaks of them are analysed,and the different peaks resulting from different vibrational models are analysed.The adsorption model on the core-shell Au/Ag nanoparticle surface is speculated.According to the characteristics of the core-shell Au/Ag nanoparticle and the mechanisms of the SERS,the different manners and the adsorption models are been speculated.From the experimental data:the different peaks in the structure of the three amino acids resulted from different vibration models;the carboxyl in the three structures all had interacted with the surface of core-shell Au/Ag nanoparticle; the different adsorption varied from the different concentrations and pH values.2.The Study of the interaction of three anticancers with DNA by SERSBleomycin,Carbonplatin and doxorubicin are selected as the probe for probing the interaction with DNA.The three anticancers' Raman spectroscopies in solid state,and the SERS on the surface of Ag nanoparticles are received and speculated;at the same conditions the SERS of the reaction of DNA with the three anticancers separately are also obtained.Through comparing the two different SERSs,each anticancer's adsorption and the effect actors on the Ag nanoparticles' surface reacted with DNA is researched,with providing the real reference for the medicine selecting.3.The Study of the Adsorption of the annular amino acids on the silver sol by SERSThe Raman spectroscopies and SERS of the three annular amino acids,phenylalanine(Phe), tryptophan(Try),histidine(His)are obtained.The different carboxyl's Raman shifts in the three molecular structures and in SERS are analyzed.On the surface of Ag nanoparticles,according to the SERS mechanisms,the adsorption of the three annular amino acids are also speculated at different concentrations and pH values.4.FT-IR,FT-Raman and Surface enhanced Raman Scattering of shikimic acidThe FT-infrared,and FT-Raman spectra of shikimic acid in solid state and in saturated liquid state are reported,which is the main material for syn anti H5N1,also have other bioactivities such as antipyretic medicine and pain-easing et al.from the molecule of shikimic acid,one carboxyl and three hydroxyl is surround the cyclohexenyl ring,this characteristic also reflected from the Raman and IR spectroscopy.All the vibrational shifts are analyzed and speculated.The adsorption of the molecule of shikimic acid on the surface of Ag nanoparticles is also obtained from the SERS and its mechanism at different concentration and pH values.All of these could provide the active conference for the developing and synthesizing new medicine with shikimic acid.5.FT-Raman study of Fmoc-amino acidsFT-Raman spectra of Fmoc-leucine,Fmoc-isoleucine,Fmoc-Metthionine,Fmoc-phenylalanine and valine are gained,the vibrational peaks are assigned.The vibrational models of Fmoc-amino acids,especially the carboxyl structure in these moleculesare speculated,which could obtain the reference information of the mechanism and vibrational model after other amino acids are combined with the Fmoc group.
