The separation of the valid ingredients of Chinese traditional medicine is the important base of the modernization of Chinese herb. Bergenin, one of the coumarin, exhibited the effect on cough, and used for codein and phlegm. Recently, foreign pharmacologists found it also have the effect of anti-AIDS and this result has aroused the field public figure's widespread interest. In this paper, bergenin was investigated by using the supercritical fluid extraction crystallization patent-technique invented by Pan Jian.(1) The content and purity of crystal were used as quality control criteria. Utilizing orthogonal test to gain an optimum extraction conditions, based on the result of single factor algorithm. The factors affecting the process were researched including temperature, pressure, time of extraction, entrainers and its adding manner. The optimized parameters are as follows: pressure 15MPa, temperature 55℃, the entrainer is ethanol (the dosage is 8%), and the result shows that the purity of crystal was more than 92% and the crystal ratio was 63.03% (The purity of material bergenin is 70%).(2) Studied the effect of different materials of crystallization board on the supercritical CO2 extraction and crystallization of bergenin, the method of crystal imprinted was introduced to improve the crystallization board, and then the process of extraction and crystallization can be enhanced. Compared with common material the imprint material has obviously effect on the crystal morphology and purity, but it has little effect on the crystal ratio.(3) The result was concluded as follows: though changing the technological conditions, the kinds of entrainer and the material of crystal board, the crystal morphology and the size of bergenin can be changed, and by doing this bioavailability of bergenin can be effected.
