Pomegranate Peel(from pishan county,xinjiang) was chosen as materials of study in this research. Extract of Pomegranate Peel was tested for its antimicrobial activity, including investigation of its antimicrobial spectrum, MIC value and its stability to different factors. Using ethanol, the extraction conditions of bioactive substances for Pomegranate Peel were optimized with orthogonal method. In addition, its application in food was also studied. The main results were as follows:1. The antimicrobial experiment was done from extract of Pomegranate Peel by petroleum ether, 60%-ethanol and water respectively. Extract of ethanol and water had antimicrobial activities and ethanol was best solvent for extracting. The optimum extraction conditions of active substances by orthogonal experiment were as follows: the ratio of material and solvent 1:30, temperature for extracting 35℃, ethanol 60%, extracting time 7h, times 3(4h+2h+1h). Under this condition, the antimicrobial diameter of extracting was 32mm.Besides, the comparison was carried on among different treatment with water. The results showed the treatment of acid and enzyme was superior to other way, treatment of alkali was lower antimicrobial activity.2. Ethanol extract of Pomegranate Peel was done for its antimicrobial properties. According to the experiments, the extract had strong inhibition to most micro-organism, namely bacteria>mould>yeast, especially to Staphylococcus aureus and Bacillus subtilis. The stronger its inhibition was with the increase of concentration and mixing time. The MIC value to Staphylococcus aureus was 3.125% and 50% to Saccharomyces cerevisiae Hansen, Baker yeast.3. Bacteria selected as sensitive micro-organism, the stability to temperature, acid, alkali, ultraviolet preserving time and metal ions were also studied. The results showed antibacterial activities were better when pH<7. The extraction was very unstable to temperature and ultraviolet when treated above 15min. Na+,Fe2+,Fe3+ could increase its stability. Fe2+, Ca2+could improve heat stability.4. After pork, bean curd, cucurbit, apricot treated by extract of Pomegranate Peel, the amount of bacteria and bacillus was much less than that of CK. Antimicrobial effect was superior to potassium sorbate to pork, bean curd, bacillus and was inferior to potassium sorbate to cucurbit, apricot. It would be very promising to be used as natural food aseptic.
