Today, people around the world focus their attention upon natural and functional material. As natural active constituents, bioactive polysaccharides and flavonoids in plant and dietary fibre play a positive role to people's health, as a functional food ingredients and food additivesit have paid people's wide attention in the world. Due to the many advantages of bamboo leaves extractions, such as abundant resource,excellent qualities,significant health function,refreshing flavour,great safety and so on, it has a wide application prospects in functional foods and healthy foods. Phyllostachys pubescens is a fine variety with the highest utilization values, and expected for the contributions to the human being. But for the whole utilizations of bamboos, the exploitation of bamboo leaves is obviously backward.Bioactive polysaccharides, flavonoids were pilot extracted from moso bamboo leaves, using biologic enzymatic technology and membrane separation technology, the resides were prepared sepecial dietary fibre of moso bamboo leaves (SDFMBL)by wet instantaneous high pressure technology ,also the physiologic activity was studied, accord with the high-efficiency,high-quality and environment-protection development trends in food field. In addition, as a polot scale production, this study can provide a reliable theoretical basis for industrialized production.1. In order to provide stabile theoretical foundation for large-scale industrialized production of polysaccharides and flavonoids from moso bamboo leaves, pilot tests by high technology were carried based on results come from laboratory. The influence of enzymes and alcohol concentration on extraction of the active constituents from moso bamboo leaves were discussed. By adding different enzymes in pilot extraction processes, influences on the extraction ratio and purification were discussed , furthermore, optimal alcohol concentration of Flavonoids extraction was studied by changing alcohol content. The test result indicate : the method of adding cellulose is the best, which could increase the extraction rates and Purities of Polysaccharides and flavonoids from moso bamboo leaves, remarkably; The optimum technical parameter for cellulose was pH was 5.0 , extracting temperature was 50℃, extracting time was 1h, the addition of pectinase was 0.15%, solid-liquid ratio was 1:15. The Optimal alcohol concentration of Flavonoids extraction was 40%.2. SDFMBL was prepared by instantaneous high-pressure from the resides using microfluidzer, the effects on water holding power,swelling,fat absorption ability, capacity of cholesterol and biliary sodium absorption and antioxidant effects on edible fat were also studied , in order to supply basis for moso bamboo leaves resources. The test results show : the water holding power and swelling increased 32% and 23% respectively; fat absorption ability for rapeseed oil and lard elevated 33.1% and 45.3%, respectively; cholesterol absorption capacities were 22.18% (pH=2) and 31.51% (pH=7) , absorption capacity power was 40% (pH=7) .Compared with AP and TBHQ, invest antioxidant effects of SDFMBL on apeseed oil and lardThe result also indicated that it had antioxidant effects on rapeseed oil and lard, and superior for the former.3. We evaluated the preventive effects of SDFMBL on hyperlipidemia and fatty liver,by feeding high fat diet adding different dosages SDFMBL, The experiment lasted for 4 weeks with blood taken from mice orbit at the final phase, and TC, TG , HDL-C and ALT in the serum are measured by automatic biochemical tester and analyzer; The raw fat in liver was measured by Soxhlet extracting and the liver cells were observed by paraffin-sections and optical microscope. The followings are the results. DFMBL could inhibit weight gain caused by high fat diet in some degree, reduce Serum Cholesterol Contents,low-density lipoprotein cholesterol, increase high-density lipoprotein cholesterol in mice obviously and repair liver injury, with the low dose has the best effects.4. We built modern mice with hyperlipidemia, fatty liver and liver damages, by feeding high fat diet, invested the lipid-reducing and liver-benifiting effect of SDFMBL to the the mice. The experiment lasted for 2 weeks with blood taken from mice orbit, and TC, TG, HDL-C and ALT in the serum are measured by automatic biochemical tester and analyzer, valued the modeling effects. Compared to normal mice, the mice with hyperlipidemia and fatty liver were divided into high lipidic group, positive control group,LSDFMBL,MSDFMBL and HSDFMBL; the experiment lasted for 2 weeks with blood taken from mice orbit, and TC, TG, HDL-C and ALT in the serum are measured by automatic biochemical tester and analyzer; The raw fat in liver was measured by Soxhlet extracting and the liver cells were observed by paraffin-sections and optical microscope. The followings are the results. SDFMBL could decrease the content of Serum Cholesterol Contents,triglyceride,low-density lipoprotein cholesterol and Alanine aminotransferase, increase high-density lipoprotein cholesterol. Reduce liver index and the content of liver crude fat, improve liver damage in mice to a certain extent, and HSDFMBL has the best effects in reducing blood lipids , LSDFMBL is the best in repairing liver function and morphological structure of the hepatocytes in mice. We invested the lipid-reducing and liver-benefiting effects of SDFMBL on morbid mice with hyperlipidemia, fatty liver and liver injury. The experiment lasted for 2 weeks with blood taken from mice orbit, and TC, TG, HDL-C and ALT in the serum are measured by automatic biochemical tester and analyzer, and detect the the modeling results.That extrating bioactive polysaccharides, flavonoids by enzymatic technology assisted with auxiliary from moso bamboo leaves and prepareing SDFMBLusing the resides is effective and feasible. SDFMBL has good water holding power,swelling,fat absorption ability, capacity of cholesterol and biliary sodium absorption,antioxidant effects on edible fat and antlipidic effect, and can provide reference for exploitation of moso bamboo leaves resources...
