Thin Layer Chromatography (TLC) is a powerful chromatography technique to separate compounds based on their differences in affinities for the mobile phases and stationary phases. TLC is a useful technique because it is relatively quick and requires small quantities of material. Thin layer chromatography scanner (TLCS) is a modern TLC instrument with multiple functions such as acquisition and storage of the spots image and quantification of the analytes. Due to the invention of TLC Scanner, TLC has become a widely used modern separation technique, distinguished by its flexibility and reliability. Although commercial TLCSs are available, their prices are usually high.In this dissertation we design a practical TLC system for TLC quantitative analysis: TLC-DC System, which use DC (Digital Camera) as detector, and a self-programmed software CIP (Chem-Image Processor) for data analysis.This TLC-DC System is simple, inexpensive and easy to use.There are six chapters in part one:In Chapter 1, the basic principles of TLC and Digital Image Processing and their applications are introduced. Firstly, TLC and its evaluation methods were discussed, especially focused on the principle of TLC Scanner and its drawback. Secondly, the application of Digital Image Processing in TLC analysis was introduced.Based on the update development in these areas, the research plan of this thesis was outlined. In chapter 2, design of TLC-System was introduced, including the consideration in light source,filter and DC, with optimized camera settings.Based on the principle of Digital Image Processing, software CIP was developed for TLC-DC System by Java language. CIP can be used in analysis of image obtained from DC, and output data into some more professional data-processing software, such as Excel, Origin, etc.. The development procedure of CIP was introduced in chapter 3. The installation and use of CIP software on Windows operation system was introduced in chapter 4.In chapter 5, a practical application of TLC-DC System was introduced: fast screening of the tetracyclines residues in fish muscle samples. The method with TLC-DC System has demonstrated to bear some requiring advantages, such as its simplicity, rapidness, low-cost, few pollutants and reasonable sensitivity.In chapter 6, the research work was summarized and the prospect of this research was given.Part TwoSolid Phase Extraction (SPE) is an extraction method that uses a solid phase and a liquid phase to isolate one, or one type, of analyte from a mixture. It is usually used to clean up a sample before separation and quantification of the analytes by using a chromatographic or other analytical method. The general procedure is to load a sample onto the SPE phase, wash away undesired components, and then wash off the desired analytes with another solvent into a collection tube.In order to speed up the procedure, accelerating the mobile phase by vacuum or by an external pressure manually is usually needed. It is difficult, however, to control the accelerating process manually. Further more, the manual accelerating operation could only carried out one column per time. In this dissertation, we design a new apparatus named Multiple-Column SPE System (MSS) to improve the efficiency of SPE.There are 4 chapters in this part:In chapter 1, the principle of SPE and the drawback of manual manipulation were presented. The design and the innovation of new system were also introduced;In chapter 2, the structure of MSS was designed and some key components of MSS were carefully compared and selected, including motor, piston, O ring, SPE column, governor and timer. A sample machine was assembled.In chapter 3, the use of MSS was described step by step. In the final chapter 4, this research work was summarized and the prospect of this research was given.
