In order to develope Penicillium sp.TS67 as an efficient biological germicide,the fermentation technology,disease-control effect and formulation process of Penicillium sp.TS67 were mainly studied.The results showed as follow:Fusarium oxysporum and Bipolaris maydis are used as indicative strains,the antifungal activity of fermented liquid from 11 antagonistic strains was determined.The results showed that strain 67 could inhibit the growth of two pathogenic fungies and the inhibiting zone diameter of 100μL fermentation liquid of strain TS67 to B.maydi was 13.6mm and to F.oxysporum was 20.3mm.The best medium for strain TS67 fermentation gained from orthogonal experiment is glucose 1%,protein peptone 0.2%,NaCl 2.5%and tween-80 0.1%.The effects of protein peptone,NaCl and glucose on antifungal material output weakened gradually.The result of single factor test indicates that the optimum fermentaion condition are 28℃,the initial pH 6.0, volume 75ml/250ml,rotation speed 170r/min,inoculation amount 8%,inoculation time 45h, culture time 96h.The output of antifungal material was enhanced reducing the rotation speed in stabilization period.Strain TS67 was grown in a 30L fermenter,the antifungal titre of 100ul TS67 fermentation liquid to B.maydis was 2735U/mg and to F.oxysporum was 3029U/mg,at 170r·min-1,28℃,8%of inoculation amount,40%of liquid volume and 1:0.9 of ventilation volume.The optimum fermentation condition of 30L fermenter is of 200r/min rotation speed and ventilation quantity 1:0.9.Through pot experiments,the disease index and control efficiency of cell,the fermentation liquid the supernatant from strain TS67 seperately to F.oxysporum and B.maydis was detected. Experiment results analysis with SPSS statistical analysis software indicated all treatments of strain TS67 could inhibit both of soybean root rot disease and corn southern leaf blight(P<0.01). Furthermore,experiment results indicated that dressing seeding with strain TS67 on soybean seed promoted soybean seeding growth.The multi-peptide antibiotic material extracted from the fermentation liquid of strain TS67 was concentrated with 60%-80%ammonium sulfate saturation.Using the concentrated fermentation liquid of strain TS67 as raw medicine,three kinds of TS67 microemulsion formulation was determined through surfactants screening.The optimum sporulation medium is sucrose 0.9499%,sodium nitrate 0.1847%,yeast extract 0.1%,artificial sea water 60%,agar 2%,distilled water 40%,pH 6.0.The storage period experiment of TS67 finish of A oil indicated that the storage period was 180 days at 4℃in dark enviroment,using conidial germination as an indicative index.The result of antifungal titre experiment of strain TS67 finishes storing 180 days at 4℃showed that it retained 3146U/mg to B.maydis and 2645U/mg to F.oxysporum and it is about 76%antifungal activity of fresh TS67 strain.
